Generating fragments from an existing antibody are commonly created by enzymatic cleavage or chemical reduction but fragments can also be generated recombinantly.

Using antibody fragments offers several advantages over full-length antibodies. Their smaller size improves tissue penetration, while the absence of the Fc region reduces non-specific binding and eliminates effector functions.

Depending on the cleavage site different fragments are obtained:

  • Above hinge region: Produces two Fab fragments
  • Below hinge region: Produces one F(ab’)2 fragment
